DNF-10® Peptide Satiety Complex (Fermented Yeast Hydrolysate from Saccharomyces cerevisiae) – 500 mg

A bioactive yeast hydrolysate clinically studied for increasing satiety, reducing caloric intake, and supporting reductions in body weight and abdominal fat.

  • Helps reduce hunger and overall caloric intake
  • Supports increased satiety between meals
  • May reduce cravings and preference for sweet foods
  • Supports reductions in body weight and body fat during calorie restriction
  • May help reduce abdominal fat while preserving lean mass

Synergy: Serves as the primary appetite-control component of the formula. Works with OEA to reinforce peripheral satiety signaling, while Caffeine, Green Tea, Cayenne, and CaloriBurn GP® support energy expenditure and thermogenesis. Importantly, the 500 mg dose used here matches the 0.5 g/day dose studied in a human trial that reduced caloric intake and body fat.

Gynostemma (Gynostemma pentaphyllum) Extract – 450 mg

A traditional adaptogenic herb rich in gypenosides that has been clinically studied for supporting AMPK activity, fat metabolism, and improvements in body composition.

  • Supports reductions in body weight and body-fat mass
  • May help reduce abdominal and visceral fat accumulation
  • Supports AMPK-mediated cellular energy metabolism
  • Promotes healthy glucose and lipid metabolism
  • May support mitochondrial function and metabolic flexibility

Synergy: Works with Forskolin, Green Tea, and CaloriBurn GP® to support multiple pathways involved in energy expenditure and fat utilization. The 450 mg dose is particularly notable because 450 mg/day has been used in randomized human trials showing improvements in body composition.

Coleus Forskohlii (Plectranthus barbatus) Extract – 250 mg

A botanical source of forskolin, a diterpene that activates adenylate cyclase and increases cyclic AMP, a cellular messenger involved in lipolysis and energy metabolism.

  • Supports cAMP-mediated metabolic signaling
  • May promote mobilization of stored fatty acids
  • Supports healthy body composition
  • May help preserve lean mass during weight-management phases
  • Complements thermogenic ingredients through a non-stimulant mechanism

Synergy: Provides a cAMP-focused pathway that complements the adrenergic effects of Caffeine and Theobromine. Works with Gynostemma and Green Tea to support fat mobilization and metabolic activity through distinct mechanisms. Human evidence for forskolin is promising but limited, and studies have not consistently demonstrated reductions in total body weight.

Green Tea (Camellia sinensis) Extract (98% Polyphenols, 50% EGCG, <1% Caffeine) – 250 mg

A highly concentrated green tea extract providing approximately 125 mg EGCG, a catechin studied for supporting fat oxidation, thermogenesis, antioxidant defense, and body composition.

  • Supports thermogenesis and daily energy expenditure
  • Promotes fatty-acid oxidation and utilization
  • May help reduce abdominal fat during weight-management programs
  • Supports healthy glucose and lipid metabolism
  • Provides potent polyphenol antioxidant activity

Synergy: Combines particularly well with Caffeine because catechins and caffeine have complementary effects on thermogenesis and fat oxidation. Works with Cayenne and CaloriBurn GP® to create a multi-pathway thermogenic complex. Human trials and meta-analyses generally find the body-weight effect to be modest rather than dramatic.

Cocoa (Theobroma cacao) Extract (99% Theobromine) – 200 mg

A highly standardized source of theobromine, a long-acting methylxanthine related to caffeine that influences adenosine signaling, phosphodiesterase activity, and metabolic pathways associated with thermogenesis.

  • Provides smooth, longer-duration methylxanthine activity
  • Supports cellular cAMP signaling
  • May promote lipolytic and thermogenic pathways
  • Supports alertness with a different stimulant profile than caffeine
  • May complement brown-fat and metabolic signaling

Synergy: Extends and broadens the stimulant profile created by Caffeine while overlapping with Forskolin at the cAMP level. Preclinical research suggests theobromine can promote white-fat browning and brown-adipose activation, but direct human evidence for fat loss or thermogenesis from isolated theobromine remains limited.

English Ivy (Hedera helix) Extract (20% Hederagenin) – 100 mg

A standardized botanical providing approximately 20 mg hederagenin, a triterpenoid being investigated for effects on AMPK, lipid metabolism, adipocyte browning, and thermogenic signaling.

  • May promote conversion of white adipocytes toward a more thermogenic phenotype
  • Supports AMPK-mediated inhibition of lipogenesis
  • May promote PKA-mediated lipolytic signaling
  • Supports healthy triglyceride and lipid metabolism
  • Provides a complementary non-stimulant metabolic pathway

Synergy: Pairs mechanistically with Gynostemma through AMPK signaling and with Caffeine, Theobromine, and CaloriBurn GP® through the broader thermogenic pathway. This is the most experimental ingredient in the formula: the anti-obesity rationale for hederagenin is currently based predominantly on cellular and animal research rather than established human weight-loss trials.

Natural Caffeine from Coffee (Coffea arabica) – 100 mg

A naturally sourced central nervous system stimulant that increases alertness while also supporting thermogenesis, energy expenditure, and fatty-acid mobilization.

  • Increases alertness, focus, and perceived energy
  • Supports acute increases in metabolic rate
  • Promotes thermogenesis and fatty-acid mobilization
  • May improve training intensity and physical performance
  • Supports energy expenditure during calorie-restricted dieting

Synergy: Works especially well with Green Tea catechins for thermogenesis and with Theobromine for a broader methylxanthine profile. Complements Cayenne and CaloriBurn GP® to amplify the formula’s energy-expenditure component. Caffeine’s appetite-suppressing effects appear relatively weak and transient, so its stronger role here is energy and thermogenesis rather than satiety.

Oleoylethanolamide (OEA) – 100 mg

A naturally occurring lipid-derived signaling molecule produced in the intestine after eating that acts primarily through PPAR-α and other pathways involved in satiety and lipid metabolism.

  • Supports natural satiety signaling after meals
  • May help reduce appetite and caloric intake
  • Activates PPAR-α pathways involved in fatty-acid metabolism
  • Supports reductions in body weight during calorie restriction
  • May improve metabolic and inflammatory markers

Synergy: Works directly alongside DNF-10® to create a dual satiety system: DNF-10® provides yeast-derived peptide signaling while OEA reinforces endogenous intestinal satiety pathways. It also complements the formula’s thermogenic ingredients by targeting food intake rather than simply energy expenditure. Human clinical research has demonstrated appetite and weight effects with OEA, although many studies have used 125–250 mg/day.

Cayenne Pepper (Capsicum annuum) Powder – 50 mg

A source of naturally occurring capsaicinoids that activate TRPV1 receptors involved in thermogenesis, fat oxidation, appetite regulation, and sympathetic nervous system activity.

  • Supports thermogenesis and energy expenditure
  • Promotes fatty-acid oxidation
  • May increase feelings of fullness
  • May reduce subsequent energy intake and food cravings
  • Supports metabolic activity through TRPV1 signaling

Synergy: Works with Caffeine, Green Tea, and CaloriBurn GP® to reinforce thermogenesis through complementary pathways. Capsaicin also adds a mild appetite-control component that complements DNF-10® and OEA. Because the label lists whole cayenne powder rather than a standardized capsaicin concentration, the actual capsaicinoid dose cannot be determined from the Supplement Facts alone.

CaloriBurn GP® Grains of Paradise (Aframomum melegueta) Extract (12.5% 6-Paradol) – 50 mg

A standardized Grains of Paradise extract providing approximately 6.25 mg 6-paradol, a pungent botanical constituent studied for activating brown adipose tissue and increasing energy expenditure.

  • Supports brown adipose tissue activation
  • Increases whole-body energy expenditure
  • Promotes thermogenic activity
  • May support reductions in visceral fat
  • Provides stimulant-free support for metabolic rate

Synergy: Complements Caffeine, Green Tea, Cayenne, and Theobromine by adding a brown-adipose-focused thermogenic mechanism without adding significant stimulant load. It pairs especially well with English Ivy’s emerging fat-browning pathway, although the human evidence base is substantially stronger for Grains of Paradise. Human research has demonstrated increased energy expenditure after Grains of Paradise ingestion.
